LU PCN is a new type of cytochrome P450 inducer. Susan West subsequently purified the cytochrome P450 from PCN-treated rats. Although electrophoretically pure, this cytochrome P450 preparation exhibited little or no catalytic activity toward a number of substrates when reconstituted with the reductase and lipid. Various attempts were made to enhance activities of the reconstituted system, but all efforts failed. As a result, we made the unwise decision to drop this project. Deuterium Isotope Effect and Metabolic Switching In 1975, Gerald Miwa joined my laboratory as a postdoctoral fellow after completing his graduate study with Arthur Cho at the University of California, Los Angeles. Because of Gerald's interest and strong background in mechanistic organic chemistry, we initiated a series of studies to investigate the mechanism of cytochrome P450-catalyzed reactions, particularly the use of a deuterium isotope as a probe. My long-term collaboration with Gerald has been scientifically and personally very rewarding. One of the most interesting studies carried out in this series was the oxidative O-deethylation of 7-ethoxycoumarin by rat cytochrome P450s 1A1 and 2B1 Miwa et al., 1984a; Harada et al., 1984 ; . Substitution of the hydrogens on the -carbon of the ethoxy side chain of 7-ethoxycoumarin resulted in an observed deuterium isotope effect of 2 for P450 1A1 and 4 for 2B1. The intrinsic isotope effects for the O-deethylation of 7-ethoxycoumarin were 1314 for both enzymes, indicating the masking of the expression of the intrinsic isotope effect by other rate factors. Careful analysis of the stoichiometric data by Nobuhiro Harada and Gerald Miwa indicated that deuterated 7-ethoxycoumarin has no effect on rat cytochrome P450 1A1 dependent NADPH oxidation, oxygen and 7-ethoxycoumarin consumption, and H2O2 formation. Thus, despite a very large intrinsic isotope effect and a significant observed isotope effect on the Odeethylation of 7-ethoxycoumarin, the oxidase and the overall monooxygenase activities of rat 1A1 are not altered. Furthermore, the decreased rate in O-deethylation was associated with a pronounced increase in 6-hydroxylation on the aromatic ring. These results suggest that 7-ethoxycoumarin can bind to the active site of rat 1A1 in at least two different orientations and that the active oxygen species, once formed, is committed to catalysis. Therefore, cytochrome P450 is capable of switching to an alternate metabolic site of the drug substrate when the preferred site of metabolism is slowed down or blocked by deuterium or other substituents. The stem cell graft.1 Two important clinical developments have evolved from identification of this immune mediated GVL effect. The first is the use of DLI to treat patients with posttransplant leukemic relapse.2-5 With DLI, the majority of patients with recurrent CML after HSCT achieve a complete remission and a smaller but significant fraction of patients with other malignancies respond to therapy.3, 6 The second is the development of allogeneic HSCT using less toxic nonmyeloablative conditioning regimens.7-12 With this approach, low doses of irradiation and chemotherapy, which alone are not sufficient to eradicate tumors, are administered to facilitate graft acceptance, and tumor regression is induced by donor immune cells. Nonmyeloablative transplants have significant activity for patients with CML, CLL, myeloma, lymphoma, and renal cell carcinoma.7-12 The potent antitumor effects observed after DLI and allogeneic HSCT in a variety of advanced malignancies represent a remarkable demonstration of the curative potential of immunotherapy in contrast to the difficulty of eliciting effective autologous antitumor immune responses to tumor associated antigens by vaccination or cellular therapy.13 However, with current approaches to allogeneic HSCT it has not been possible to separate the beneficial GVL effect from deleterious GVHD. Effector Mechanisms in the Graft-versus-Leukemia Response A more complete understanding of the cellular mechanisms operative in an allogeneic GVL response should provide critical insights into the requirements for effective antitumor immunity and potentially permit separation of GVL from GVHD. The association of GVL activity with GVHD has implicated donor T cells reacting with minor histocompatibility H ; antigens expressed by recipient cells as major contributors to the GVL effect. The first clinical demonstration of GVL activity was observed after allogeneic HSCT for advanced leukemia in which the probability of leukemic relapse was found to be significantly lower in those patients who developed acute and or chronic GVHD.14, 15 Analysis of patients with CML, AML, or ALL treated with either allogeneic unmodified HSCT, allogeneic T cell depleted HSCT, or syngeneic HSCT showed that the risk of relapse was lowest for patients who received allogeneic unmodified HSCT and developed acute and or chronic GVHD.1, 16 Transplantation with syngeneic or T cell depleted allogeneic marrow to avoid GVHD was associated with a higher risk of relapse unless the conditioning regimen was intensified.1, 17, 18 However, GVHD is not a prerequisite for GVL activity. The subsequent implantation rate per transferred embryo also did not differ between the three groups: 16.7% in MB2, 12.2% in Medi-Cult and 11.9% in the mixed group 2; NS ; . Study II MB2 versus BM1 In 117 consecutive oocyte retrievals from patients undergoing infertility treatment by IVF, 1518 oocytes were retrieved, i.e. a mean SD of 12.9 8.1 per oocyte retrieval. About half of the OCCs n 765 ; were cultured in MB2 medium and the remaining sibling OCCs n 753 ; in BM1 medium. The results are shown in Table IV. In the same cohort of oocytes, a similar fertilization rate was obtained for the oocytes cultured in MB2 66.9 27.5% ; as for those cultured in BM1 62.0 31.7% ; paired t-test; NS ; . In both media, the proportion of abnormal fertilizations was similar; 5.5% of the inseminated oocytes showed one pronucleus and 4.4% were polyspermic in MB2 versus 5.3% with 1PN and 4.2% with 3PN in the BM1 medium paired t-test; NS ; . The total cleavage rates were very similar; in MB2 84.1% of the zygotes developed into transferable embryos and in Medi-Cult 81.0%. No differences between the proportions of type A, B and C embryos were observed between the two media. The stage of embryo development at the moment of the first evaluation 41 h after insemination ; was significantly different between the two groups. A significantly greater proportion of embryos had reached at least the three- or four-cell stage after culture in MB2 P 0.001 ; than after culture in BM1. For transfer, the best embryos in terms of morphological appearance and rate of development were selected, irrespective of the medium. In three 2.5% ; out of the 117 oocyte retrievals, a transfer could not be carried out owing to fertilization failure n 1; 0.9% ; or to a low fertilization rate followed by bad embryo development n 2; 1.7% ; . Of the 114 transfers, 27 were solely of embryos cultured in MB2 group I ; , eight were solely of embryos cultured in BM1 group II ; and the remainder n 79 ; were of embryos cultured in either one or the other medium mixed group ; . The data on the transfers and the outcome of the transfers are given in Table V. The mean age of the patients was significantly lower in the MB2 group 30.8 years ; than in the MB1 and the mixed group 33.4 years ; . The number of embryos transferred was significantly different.

Experiments are under way to determine whether UO-44 gene promoter contains AP-1 recognition site s ; and its expression is regulated by the products of the above protooncogenes in response to tamoxifen and estrogen treatment. The common mechanism by which tamoxifen, GH, and estradiol regulate UO-44 gene expression in the rat uterus is not well understood. It has been reported that GH, tamoxifen, and estrogens have similar effects on UO-44 and IGF-I expression in the uterus, whereas ICI 182780 inhibits it 16, 44 ; . This is consistent with studies in experimental animals showing that these compounds produce similar effects on uterine weight. It is possible that the effect of these compounds on UO-44 expression involves IGF-I as a mediator. Experiments are under way to determine this possibility.
